Complex proteins at lightning speed: GPEx® Lightning™ and a 10-month path to drug substance

Bispecific, multispecific, and other multichain molecules have traditionally been among the most challenging modalities to develop, often requiring longer timelines due to their complexity. Chain mispairing, unbalanced expression, aggregation and difficult purification push these programs onto longer, less predictable paths than conventional monoclonal antibodies, often delaying critical clinical decisions by many months.

Complex molecules no longer serve as a barrier to speed to the clinic. This session examines the technical foundations behind a defined 10-month path from gene sequence to released GMP drug substance for eligible complex molecules. The discussion centers on GPEx® Lightning cell line development, where dock-directed integration into transcriptionally active regions supports stable pools in approximately 25 days and titers exceeding 11 g/L for bispecific antibodies, with stability demonstrated across more than 60 generations and taken off the critical path.

The presentation will detail how chain-ratio selection is executed on pool material through small-scale expansion and platform analysis, allowing the process to be locked early and downstream, analytical and formulation development to proceed in parallel rather than sequentially. Attendees will see how parallel execution and early representative material compress the critical path while maintaining quality and scalability, and which molecule characteristics determine whether a program fits an accelerated path.
